We have applied non-negative matrix factorization to a database of leaf hyperspectral reflectance and DMSO chlorophyll extract absorption measurements from grapevine canopies at seven vineyards in Dalmatia, Croatia. Extracted spectral signatures show various monthly changes in grapevine production of chlorophyll. Our signatures represent maximal absorption values within a specific colour spectrum - blue, red, near infrared, green and yellow to orange. Association of signatures and chlorophyll indices vary through time. Signature 4 is the best chlorophyll proxy. Here we show that the same amount of chlorophyll can be produced by plants using multiple internal processes or absorption of different spectrums of light. Changes in these processes can be better understood by studying extracted reflection signatures instead of just chlorophyll concentration or vegetation indices. This study shows that non-negative matrix factorization applied to hyperspectral reflectance measurements can be a powerful tool in monitoring the short-term changes in physiology of plants thus could be applied in precision viticulture. We also tested this model on canopy hyperspectral reflectance measured at 0.5 m distance from the canopy. This study shows that NMF can be a powerful tool in monitoring the seasonal grapevine changes in physiology of plants thus could be applied in precision viticulture.

SUMMARYThe abandonment of Asturian vineyards over the last century almost resulted in the extinction of the grapevine crop and major cultivar confusion. Due to the restoration of a number of these vineyards in recent years, research into the regional varietal stock is needed. The aims of the present study were to characterise genetic resources of Vitis vinifera L. in Asturias and to identify synonyms and homonyms through an analysis of six microsatellite markers: VVS2, VVMD5, VVMD7, VVMD27, VrZAG62, and VrZAG79. These markers are used internationally as recommended descriptors for the identification of grapevine cultivars and allow comparisons between laboratories. After PCR amplification of these microsatellite sequences, they were analysed by capillary electrophoresis. Forty-six accessions of V. vinifera L., corresponding to 14 cultivars, were analysed, but only nine different genotype profiles were found, due to synonyms for 'Albarín Tinto', 'Albarín Blanco' and 'Verdejo Tinto', and two homonyms for 'Albarín Blanco' being identified ('Godello' and 'Savagnin Blanc'). The most informative locus was VVMD5 and the least informative was VVMD27. The heterozygosity observed at all loci was higher than expected.

Aim: The objective of the present study is to investigate the molecular characteristics of Sardinian grapevine cultivars to evaluate cases of synonyms and false attributions to protect local agro-biodiversity.Methods and results: The SSR analysis (13 loci) has been used to define the DNA fingerprint and the relationships with Sardinian grapevine cultivars. Results highlighted a high genetic variability among the accessions, with the Dice coefficients performing from 0 to 0.8. Despite the genetic richness, thirteen groups of redundant genotypes were detected. Molecular analysis refers of cultivars harbouring the same SSR profile but different berry colours such as cultivars Licronaxu Bianco and Nero and Moscatello Bianco and Nero. It could by hypothesized that Licronaxu and Moscatello could derive from a specific retrotransposon-induced mutation event in genes regulating anthocyanin biosynthesis.Conclusion: Sardinian germplasm has a real problem of cultivar identification probably due to different factors such as the absence of an exhaustive ampelography, problems in the language to name varieties and the existence of cultivars sensitive to biotic and abiotic stresses producing evident morphological modifications leading to mistakes in recognising and identifying properly the affected plants. However, our molecular results suggest that high grape-biodiversity is still preserved in this region.Significance and impact of study: Results of this work clarified the relationships among grapevine cultivars and provided a solid basis to improve a regional grapevine collection.
