“…During the intracellular reduction process of CrVI to CrIII, a spectrum of reactive oxygen species (ROS) (ie, superoxide [O2•À], hydrogen peroxide [H2O2], and hydroxyl radical [•OH]) is produced, with the depletion of antioxidant enzymes culminating in increased oxidative stress (Standeven and Wetterhahn, 1991). We have identified oxidative stress as one of the major pathways for CrVI-induced male (Kumar et al, 2017) and female reproductive toxicity (Banu et al, 2016;Stanley et al, 2013Stanley et al, , 2014) in a rat model. However, there is no report on the impact of Cr on placental oxidative stress in the human.…”

“…Chromium exposure has been associated with these pathological changes which may be a cause of impaired spermatogenesis. Additionally, chromium can cross the blood-testis barrier and effect sperm development, maturation, and sperm motility (Kumar et al 2017). We observed that sperm motility was more affected than any other parameters.…”

“…Gut Histology: Cryosections of the ileum were stained for Hematoxylin and Eosin (H&E) as previously described (Kumar et al, 2017). Sections were visualized and photographed with the FSX100 Cell Imaging System (Olympus) at 10X magnification.…”

“…Immunohistochemistry: Immunofluorescence for Zonula Occludens (ZO-1) was performed as previously described (Kumar et al, 2017). Briefly, a portion of the ileum was embedded in Cryo-OCT compound (Leica Microsystems, Buffalo Grove, IL, USA) and frozen in liquid nitrogen.…”
