MALDI-TOF mass spectrometry confirms difficulties in separating species of theAvibacteriumgenus

Abstract: In the present study a well-characterized strain collection (n = 33) of Avibacterium species was investigated by matrix-assisted laser desorption ionization-time-of flight mass spectrometry (MALDI-TOF MS). The robustness of the currently available reference database (Bruker Biotyper 3.0) was tested to determine the degree of identification of these strains. Reproducible signal patterns were obtained from all strains. However, identification of most strains was only possible at genus level. Furthermore, two str… Show more

“…Misidentification by MALDI-TOF MS has been associated with insufficient numbers of reference strains within the database (25). The difficulties of confident species identification by MALDI-TOF MS reported in the current case are similar to those reported for another genus of the family Pasteurellaceae, Avibacterium (26). The failure of the MicroSeq system to identify the isolate was essentially due to the absence of M. glucosida from the database.…”

“…The close‐similarity among MALDI‐TOF profiles of main fish pathogenic species of Aeromonas was reflected by 2D peak distribution diagram (Figure ) showing an overlapping among isolates of different species. This overlapping is likely related with the strong phylogenetic relatedness within the genus Aeromonas (Martin‐Carnahan & Joseph, ) and in line the limited discrimination power of MALDI Biotyper to separate closely related species observed in other microorganisms (Alispahic, Christensen, Bisgaard, Hess & Hess, ; Frey & Kuhnert, ).…”

“…MALDI‐TOF‐MS platform has been recently tried on some rare bacteria such as Avibacterium, Myroides, Granulicatella , and Abiotrophia , with comparable results and advantages . In addition, recent reports have shown that both identification and antibiotics susceptibility test results can be obtained together in a few hours from positive blood culture by MALDI‐TOF‐MS .…”
