MALDI–MS Profiling to Address Honey Bee Health Status under Bacterial Challenge through Computational Modeling

Abstract: Honey bees play a critical role in the maintenance of plant biodiversity and sustainability of food webs. In the past few decades, bees have been subjected to biotic and abiotic threats causing various colony disorders. Therefore, monitoring solutions to help beekeepers to improve bee health are necessary. Matrix‐assisted laser desorption ionization–mass spectrometry (MALDI–MS) profiling has emerged within this decade as a powerful tool to identify in routine micro‐organisms and is currently used in real‐time … Show more

“…Gut tissues were washed in PBS and then placed in low protein binding tubes (LoBind, Eppendorf, France) containing 20 μl of TFA 1% and vortexed. Hemolymph was collected from the same 12 bees according to the procedure published by Arafah and colleagues [69].…”

“…This approach which has been greatly improved over the past 20 years, has demonstrated its feasibility, robustness and cost-effectiveness in identifying a wide range of microorganisms (e.g., bacteria, mycobacteria and certain fungal pathogens [70], parasites [71,72] and more recently some microsporidia pathogenic for honeybee [73]. We also used MALDI MFPs to monitor the impact of experimental microbial infections in honeybee hemolymph [69]. To create model MFPs, an average spectrum for each tissue was recorded as the most representative histomolecular signature of the tissues (Figure S1).…”

“…Among these results, the work of Arafah et al [ 231 ] proposed and developed MALDI–MS profiling to address honeybee health status under bacterial challenge through computational modeling, while Edwards and Dixon [ 232 ], in 2016 published a paper regarding negotiating the conflict between humans and honeybees towards an ecological city in the journal of Society and Animals .…”

“…Using modified OECD risk assessment protocols and fully crossed experiments, combined with MALDI BeeTyping ® and bottom-up proteomics by LC-ESI-MS/MS [ 22 , 63 , 64 ], we ask (i) how exposure to an insecticide, fungicide, and herbicide, individually or in combination with the parasite, impacts the haemolymph proteome profile; (ii) which proteins respond to these stressors; and (iii) how these responses map onto higher level effects of exposure on individual longevity, weight change, and parasite load.…”
