MALDI-Mass Spectrometry Imaging to Investigate Lipid and Bile Acid Modifications Caused by Lentil Extract Used as a Potential Hypocholesterolemic Treatment

Abstract: This paper reports matrix-assisted laser desorption/ionization mass spectrometry imaging to investigate systematic effects of a lentil extract treatment to lower cholesterol levels. For this purpose, mass spectrometry imaging was used to spatially investigate modifications in the lipid composition and cholesterol levels in the brain, liver, and intestines as well as bile acids in the liver and intestine of rats treated with lentil extract. Neither the lipid composition nor cholesterol levels in the brain sampl… Show more

“…Regarding cholesterol level, we found a strong increase in its concentration, even though not reaching statistical significance in CAF rats. It is well known that rodents are resistant to develop hypercholesterolemia [ 69 , 70 , 71 , 72 ], unless longer exposure to HFD or specific cholesterol-enriched diets [ 73 , 74 ]. However, the probiotic supplementation showed a clear trend toward a reduction of cholesterol, triglycerides and a significant decrease in glycemia and LDL levels in CAF+P rats.…”

“…Most imaging experiments have probed discrete regions of the intestine for analysis due to the difficulty in preparing large sections of intestine for sectioning (Carter et al, 2019). Our study, however, underscores the importance of including luminal content in MALDI IMS analysis for a more representative analysis of the gut, as seen by the bile acid localization patterns previously not shown with traditional sample preparation approaches (Figure S1) (Genangeli et al, 2019).…”

“…Beta-carboline, commonly referred to as norharmane (C 11 H 8 N 2 , 168.06875 u), is a nitrogen-containing heterocycle used for the analysis of lipids in both polarities 39 as well as bile acids in the negative ion mode. 40 In the negative ion mode, identified clusters do not exceed m/z 670 and are mainly of the [aM-H-bH 2 ] À…”
