Malaria parasite centrins assemble by Ca2+-inducible condensation

Abstract: Centrins are universally involved in eukaryotic cell division, but their precise mode of function remains unclear. They are small calcium-binding proteins and a conserved component of microtubule organizing centers (MTOCs) that organize the mitotic spindles. Malaria-causing parasites have a particularly divergent acentriolar MTOC, which incorporates several essential centrins. Here, we reveal calcium-inducible liquid-liquid phase separation as a principle of assembly for Plasmodium and human centrins. We defin… Show more

“…22.533773 doi: bioRxiv preprint MTOC and microtubules during the trophozoite and schizont stages by pairing NHS ester, which we have recently shown can stain both the intranuclear and cytoplasmic portions of the MTOC 6 , with an anti-centrin antibody (Clone 20H5, raised against centrin from Chlamydomonas). This antibody likely recognizes centrin 3 in P. falciparum 20 , but a recent study suggests that all four P. falciparum centrins share an MTOC localization 21 .…”
