Major-surface-protein-4-gene-based detection of Anaplasma marginale isolated from sheep in Al-Diwaniyah province, Iraq

Abstract: This study was purposed for confirming detection and typing of Anaplasma spp in infected sheep from Al-Diwaniyah province, Iraq. Sampling of 50 blood specimens was performed from clinically-identified infection of anaplasmosis. The samples of the blood were subjected to DNA extraction followed by polymerase-chain-reaction (PCR)-based detection of the Anaplasma marginale using major surface protein (MSP4) gene. The results have shown that 8 blood samples were infected with A. marginale. The PCR-based identifica… Show more

“…For the PCR reaction, the steps were denaturation at 94°C for 4 min, annealing at 63.1°C for 45 sec, elongation at 72°C for 45 sec, and a final extension at 72°C for 5 min. A 2 % agarose gel was used to visualize PCR products (15).…”

“…Once liberated from the parasitized leukocytes, merozoites invade erythrocytes, where they grow into piroplasms (17)(18)(19)(20)(21)(22). Conjunctival petechial hemorrhage, swollen lymph nodes, and anemia are symptoms of theileriosis, in addition to high fever, restricted appetite, loss of body weight, and general weakness (23,24). Theileria piroplasms are often seen in animals and acute sings and serve as reservoirs for the parasite community (25).…”
