Maize Urb2 protein is required for kernel development and vegetative growth by affecting pre‐ribosomal RNA processing

Abstract: Ribosome biogenesis is a fundamental process in eukaryotic cells. Although Urb2 protein has been implicated in ribosome biogenesis in yeast, the Urb2 domain is loosely conserved between plants and yeast, and the function of Urb2 protein in plants remains unknown. Here, we isolated a maize mutant, designated as urb2, with defects in kernel development and vegetative growth. Positional cloning and transgenic analysis revealed that urb2 encodes an Urb2 domain-containing protein. Compared with the wild-type (WT), … Show more

“…For scanning electron microscopy (SEM), mature kernels were prepared according to Lending & Larkins (1992), and observed under a scanning electron microscope (S3400N; Hitachi, Tokyo, Japan). For transmission electron microscopy (TEM), 21 d after pollination (DAP) kernels were prepared according to Wang et al ., (2018), and observed under a transmission electron microscope (H7600; Hitachi).…”

“…The ZmDRH1 protein localizes to the nucleolus and interacts with the RNA binding protein MA6 and FIBRILLARIN (Gendra et al, 2004). The others, UNHEALTHY RIBOSOME BIOGENESIS2 (ZmURB2) and RIBOSOME EXPORT ASSOCIATED1(ZmREAS1) are required for kernel development and are involved in pre-rRNA processing and ribosome assembly (Qi et al, 2016;Wang et al, 2018).…”

“…Several important intracellular biological processes, such as post-transcriptional regulation, also play important roles in maize kernel development. These processes have been uncovered by studying mutants rbm48 (Bai et al, 2019), ubl1 (Li et al, 2017b), and urb2 (Wang et al, 2018). These findings have enriched our knowledge of the regulatory mechanisms behind maize kernel development.…”
