Maize stripe virus RNA5 is of negative polarity and encodes a highly basic protein

Abstract: The entire 1317 nucleotide sequence of the maize stripe virus (MStV) RNA5 was determined. Only one open reading frame (ORF) was identified and was found in the viral complementary RNA (vcRNA). This ORF appears to encode a protein ofM r 44237, hereafter referred to as NS 5. In vitro translation of transcripts representing nearly full-length RNA5 vcRNA yielded products of the predicted size, as well as some smaller, less prominent products. No products were identified from transcripts of viral polarity. RNA hybr… Show more

“…Each of RNAs 2, 3 and 4 has an ambisense coding strategy, and RNA segment 1 is a negative strand RNA. MStV, another member of the tenuiviruses, contains five RNA segments, of which the smallest RNA (RNA 5) is a negative strand and encodes a highly basic protein (Huiet et al, 1993). Although the existence of a small RNA has been reported for a different isolate of RSV (Ishikawa et al, 1989), we have been unable to find such a small distinct RNA in RSV (Toriyama, 1982;Toriyama & Watanabe, 1989).…”

“…We obtained an RNA sample originating from the genomic sequencing and characterization work previously performed with a Florida, United States of America (USA) isolate of MSpV (Falk and Tsai, 1984;Huiet et al, 1991Huiet et al, , 1992Huiet et al, , 1993Estabrook et al, 1996). We refer to this isolate as MSpV21.…”

“…The genome of a Florida (United States of America) isolate of MSpV was composed of five RNAs (Falk and Tsai, 1984), and the complete sequences for RNAs 2-5 were determined (Huiet et al, 1991(Huiet et al, , 1992(Huiet et al, , 1993Estabrook et al, 1996). In a separate effort, a partial sequence of RNA1 from an isolate of MSpV from Réunion (France) was determined (Mahmoud et al, 2007).…”

“…Note added in proof It has been recently reported that the 5' ends of the MStV mRNA4 population contain 10 to 15 additional nucleotides and that they are heterogeneous in sequence [Huiet et al (1993) Virology 197,[808][809][810][811][812].…”

“…The nucleotide sequence of the RNA5 segment of MStV (Huiet et al, 1993) has been determined. An RNA5 species was also observed in one isolate of RSV (Ishikawa et al, 1989).…”
