Maintenance of the Specification of the Anterior Definitive Endoderm and Forebrain Depends on the Axial Mesendoderm: A Study Using HNF3β/Foxa2 Conditional Mutants

Abstract: In mouse embryo, the early induction of the head region depends on signals from the anterior visceral endoderm (AVE) and the anterior primitive streak. Subsequently, node derivatives, including anterior definitive endoderm and axial mesendoderm, are thought to play a role in the maintenance and elaboration of anterior neural character. Foxa2 encodes a winged-helix transcription factor expressed in signaling centers required for head development, including the AVE, anterior primitive streak, anterior definitive… Show more

“…However, defects in ANP development were not fully penetrant and therefore could be secondary: Recent studies point to a role of node derivatives such as the NC and somites in maintaining and stabilizing anterior neural specification (53). In Hnf3b/Foxa2 conditional mutants, which lack paraxial mesoderm, specification of the ANP occurs, but is labile (54).…”

“…Embryos in which extra-embryonic lineages are wild-type while the epiblast lacks Foxa2 fail to form a node and notochord, have fused somites, and establish rudimentary neural patterning (Dufort et al, 1998;Hallonet et al, 2002). Foxh1 mutant embryos also lack APS derivatives, and show somite fusion (Hoodless et al, 2001;Yamamoto et al, 2001).…”

“…This is due, in part, to the relative lack of genetic markers of this tissue. Many genes that are expressed in the definitive endoderm, such as Hex, Cer1, Foxa2, and Sox17, are also expressed in the extraembryonic visceral endoderm (Ang et al, 1993;Belo et al, 1997;Biben et al, 1998;Shawlot et al, 1998;Thomas et al, 1998;Hallonet et al, 2002;KanaiAzuma et al, 2002). A second limitation of these definitive endoderm markers is that they are not expressed in the entire definitive endoderm at any point during gastrulation but instead only mark specific regions of the definitive endoderm.…”
