Maintenance of Normal in situ Chromosomal Features in Long-Term Tissue Cultures

Abstract: Clonal isolates from a rapidly proliferating fibroblast-like derivative have retained the classic diploid chromosome relationship over a period of many transplant generations. The readily identified members of the 11 pairs of chromosomes, including the sex chromosomes (X(1) and X(2)), have aided in localizing minute structural alterations within recloned diploid and aneuploid sublines.

“…Between 4 and 6 million CH cells, the equivalent of 0.5-1 confluent 10-cm Petri dish, were dissociated with trypsin, resuspended in Ϸ0.3 ml of PBS, and injected into in the upper hind leg of 6-9-month-old syngeneic, male CHs (34). Tumors of 1-1.5 cm were explanted at the time of diagnosis, minced with scalpels, dissociated into single cells with trypsin, and then cultured until a confluent Petri dish was obtained for the analysis of metaphases as described (24).…”

“…Subconfluent cultures of CHE cells, derived from a single male, 18-day embryo (71), that had been kept growing for one or two consecutive passages in culture were incubated for 2-3 hr with 0.1 g͞ml of colcemid (GIBCO͞BRL). The cells were then rinsed with phosphate-buffered physiological saline, dissociated with trypsin at 37°C, mixed with 1 ml complete culture medium, and centrifuged for 6 min at 500 rpm at room temperature.…”

“…The CHE in vitro system also allows a distinction between chemical and spontaneous transformation, because spontaneous transformation occurs at a much lower frequency and, on average, only many more cell divisions later (19,(71)(72)(73) than chemical transformation (20,66,68). We have also studied the chromosomes of spontaneously transformed CHE cells.…”
