Magnesium Lithospermate B Protects Neurons Against Amyloid β (1–42)-Induced Neurotoxicity Through the NF-κB Pathway

Abstract: Magnesium lithospermate B (MLB) is one of the major bioactive components of Radix Salviae miltiorrhizae, a Chinese traditional herbal medicine colloquially known as Dan Shen. In this study, we investigated the neuroprotective effect of MLB against oligomeric amyloid β (Aβ) (1-42)-induced neurotoxicity in cultured FVB mouse hippocampal neurons. We found that pretreatment with MLB not only prevents a loss in neuronal cell viability following exposure to Aβ (1-42), but also attenuates Aβ (1-42)-induced release of… Show more

“…Our study found that SH-SY5Y cells exposed to Aβ displayed higher concentration and expression of IL-1β, TNF-α and IL-6 than that of the control groups. In agreement with our results, Jiang et al [25] revealed that Aβ could induce release of pro-inflammatory cytokines IL-1β, IL-10, and TNF-α. Previous studies confirmed that PGE2 played pivotal functions in inflammation and was elevated together with the expression of COX-2, a rate-limiting enzyme catalyzing PGE2 production, in the brain of AD patients [22, 23].…”

“…The NF-κB pathway is well known to promote the expression of genes related to neuronal inflammation [18][19][20]. Our cumulative results demonstrated the anti-inflamma- Accumulation and aggregation of Aβ peptides generally lead to activation of neuroglia cells, which eventually initiates neuronal oxygen response and release of inflammatory cytokines such as interleukin-1β (IL-1β), IL-6, and tumor necrosis factor-α (TNF-α) [16,17].…”

“…The NF κ B pathway has been shown to be involved in the pathological effects of A β [ 42 44 ]. It has also been reported that the anti-inflammatory effects of statins work at least in part through inhibition of the NF κ B pathway [ 45 47 ].…”
