Mad2 promotes Cyclin B2 recruitment to the kinetochore for guiding accurate mitotic checkpoint

Abstract: Accurate mitotic progression relies on the dynamic phosphorylation of multiple substrates by key mitotic kinases. Cyclin‐dependent kinase 1 is a master kinase that coordinates mitotic progression and requires its regulatory subunit Cyclin B to ensure full kinase activity and substrate specificity. The function of Cyclin B2, which is a closely related family member of Cyclin B1, remains largely elusive. Here, we show that Mad2 promotes the kinetochore localization of Cyclin B2 and that their interaction at the … Show more

“…In most of the cases reported, poor checkpoint regulation due to overexpression of specific cyclins or spindle assembly factors were the cause for the appearance of mitotic slippage and subsequent tetraploidization [91,[93][94][95][96]. Our results show that RNF144B could be involved in regulating the levels of several cell cycle and spindle assembly proteins, including TPX2 [79], MAP4 [97], BCCIP [98], CCNB2 [99], RAD21 [81] and KIF4 [100].…”

“…CDK2 is a positive regulator of cell proliferation, and cell differentiation depends on its decreased activity [ 22 ]. In eukaryotic cells, CCNB1 and CCNB2 are cyclins which regulate the cell cycle and are closely related functionally [ 23 ]. Overexpression of CCB1 effectively induces cell division in post-mitotic mouse, rat and human cardiomyocytes [ 24 ].…”

“…The DNA was stained with 4′,6-diamidino-2-phenylindole (DAPI) from Sigma. Images were captured by DeltaVision softWoRx software (Applied Precision) and processed by deconvolution and z-stack projection ( Liu et al., 2022 ). Images were deconvolved using SoftWoRx (Applied Precision) and processed with ImageJ.…”
