Macrophage deactivation by interleukin 10.

Abstract: Recombinant mouse interleukin 10 (IL-10) was exceedingly potent at suppressing the ability of mouse peritoneal macrophages (m phi) to release tumor necrosis factor alpha (TNF-alpha). The IC50 of IL-10 for the suppression of TNF-alpha release induced by 0.5 microgram/ml lipopolysaccharide was 0.04 +/- 0.03 U/ml, with as little as 1 U/ml suppressing TNF-alpha production by a factor of 21.4 +/- 2.5. At 10 U/ml, IL-10 markedly suppressed m phi release of reactive oxygen intermediates (ROI) (IC50 3.7 +/- 1.8 U/ml),… Show more

“…The amount of NO produced by the METH extract (105.99 µmol/5 × 10 5 cells) was larger than the production of the DCM extract (58.9 µmol/5 × 10 5 cells) (Figure 1). The results regarding TNF-α confirmed a significant production of this cytokine, at levels near that of the positive control (252.7 and 234.6 units/ mL, METH and DCM extracts, respectively), confirming a correlation between the synthesis of TNF-α and NO (Figure 2) (Bogdan et al, 1991;Carli et al, 2009). I. suffruticosa did not produce significant amounts of H 2 O 2 when compared with the negative control (p > 0.05): METH, 0.59 nmol/2 × 10 5 cells and DCM, 3.3 nmol/2 × 10 5 cells) (Figure 3).…”

“…The IL-10 activity minimizes the tissular impairment occurred in the disease's site through inhibiting the production of proinflammatory cytokines (Bogdan et al 1991, D'Andrea et al 1993. In this study, PBMCs of patients with active pulmonary TB were observed to produce high levels of IL-10.…”

“…9, 41 We believe that vIL-10 not only deactivates macrophages via the prevention of IFN-g production by Th1 cells, but also deactivates macrophages directly, and inhibits the production of TNF-a by macrophages. MCP-1 is released from many types of cells, including macrophages, lymphocytes, platelets, fibroblasts, endothelial cells, epithelial cells, 42 and mesangial cells.…”
