Macrophage CD74 contributes to MIF-induced pulmonary inflammation

Abstract: MIF-induced neutrophil accumulation in the alveolar space results from interaction with CD74 expressed on the surface of alveolar macrophage cells. This interaction induces p44/p42 MAPK activation and chemokine release. The data suggest that MIF and its receptor, CD74, may be useful targets to reduce neutrophilic lung inflammation, and acute lung injury.

“…Myosin light-chain kinase, a central cytoskeletal regulator encoded by Mylk, has a key pathophysiological role in ALI (26); and v) Mif, macrophage migration inhibitory factor, is involved in the pathogenesis of several inflammatory diseases. Mif-induced neutrophils accumulate in the alveolar space, indicating that Mif may be a useful target in the reduction of neutrophil lung inflammation and ALI (27). These methylated genes are involved in important mechanisms that underlie ALI/ARDS.…”

“…ICAM-1 is involved in neutrophil chemotaxis by binding with the surface ligand CD11/CD18 on neutrophils and lymphocytes (Dominguez et al, 2015;Liu et al, 2015;Sumagin and Parkos, 2015). MIP-2 belongs to the chemokine C-X-C subfamily and induces marked chemotaxis in neutrophils and lymphocytes (Takahashi et al, 2009;Tateno et al, 2013;Vagima et al, 2015). ALI occurrence was closely associated with neutrophil and lymphocyte accumulation in lung tissue.…”

“…4,5 However, increasing use of fluorescent molecules from a wide range of emission (em) wavelengths (400 to 700 nm) needs MWSM with a broader fluorescence spectrum. 6,7 Currently only a few numbers of MWSMs with limited wavelengths are known and commercially available to date. 8 For example, GE Healthcare's latest MWSM of dual indication with visible and fluorescent dyes is useful for proteomics analysis, but the use of Cy3 and Cy5 dyes limits it detection range.…”
