Macrolide antibiotics as antiinflammatory agents: Roxithromycin in an unexpected role

Abstract: The antiinflammatory activity of a new 14-membered macrolide antibiotic, roxithromycin, was evaluated in various rat models including carrageenan- and poly-L-arginine-induced hind-paw oedema, croton oil inflamed ear assay and polyester sponge granuloma. When administered orally to animals, roxithromycin displayed an atypical profile in the assays utilized, including: (1) marked antioedema activity similar to that of indomethacin in poly-L-arginine assay, (2) significant inhibition of lambda-carrageenan hind-pa… Show more

“…6 . Also, macrolides exhibited less anti-inflammatory activity than ibuprofen; this is similar to the findings of Agen C et al 19 . Change in paw volume in the combination groups (Ibuprofen 100 mg/kg + Roxithromycin 20 mg/kg and Ibuprofen 100 mg/kg + Erythromycin 20 mg/kg) was not significantly different from that of Ibuprofen (p> 0.05); hence it appears that there is no additional anti-inflammatory activity in the combination groups.…”

“…Macrolides have been shown to affect several pathways of the inflammatory processes such as the migration of neutrophils, the oxidative stress in phagocytes and the production of proinflammatory cytokines. 9 The anti-inflammatory activity of roxithromycin have been shown to be due to reduce production of nitric oxide (NO) and super oxide dismutase activity (Agen c. et. Al, 1993).…”

“…Patients with cystic fibrosis experienced clinical improvement and fewer exacerbations on maintenance azithromycin therapy (138)(139)(140). Macrolide antibiotics exert both antiinflammatory and nonbactericidal antimicrobial effects (141)(142)(143). In a retrospective analysis of more than 200 lung transplant recipients, Johnson and colleagues (144) demonstrated a strong association between the use of statins to control hypercholesterolemia, and freedom from the development of BOS.…”
