m6A RNA methylation regulators could contribute to the occurrence of chronic obstructive pulmonary disease

Abstract: Chronic obstructive pulmonary disease (COPD) is characterized by a progressive condition of small airway obstruction, chronic bronchitis and emphysema, representing a leading cause of death worldwide. 1 Although oxidative stress, immunity and inflammation, apoptosis, metaplastic epithelial lesions, mucus hypersecretion and fibrosis in both airways and lungs are hallmarks of this disease, 1,2 the exact pathogenesis of COPD remains unclear. To date, more than 100 kinds of RNA modifications have been identified i… Show more

“…Certain studies have reported that METTL3 contributes to transforming growth factor-β-induced epithelial-mesenchymal transition of lung cancer cells (39). An additional study revealed that there is a significant correlation between METTL3 expression and the occurrence of chronic obstructive pulmonary disease (40). However, the role of METTL3-mediated m6A modification in NSCLC initiation and progression is not fully known.…”

“…Subsequently, the connectivity per gene was deduced through calculating connection strengths with other genes. After validating the module structure preservation using the module preservation R function, the gene expression profile of each module was summarized by the module eigengene on whom IPF traits were regressed in the Limma R package ( Huang et al, 2020 ). The IPF-related module was selected with the highest coefficient square (r 2 ) and the p value <0.001.…”

“…The function of m6A-modified targets is executed by m6A binding proteins (readers) through binding to m6A directly or indirectly, including YTH N6-methyladenosine RNA-binding proteins, YTH domain-containing proteins, IGF2BPs, and HNRNP protein families ( Tong et al, 2018 ). These writers, erasers, and readers, serving as m6A RNA methylation regulators, can influence mRNA of target gene at different levels, including nuclear export, translation, splicing, stability, and decay ( Lan et al, 2019 ; Huang et al, 2020 ; Nombela et al, 2021 ) and have important influences in diverse physiological and pathological processes, such as cell proliferation and differentiation, oncogenic protein expression, and the proliferation, survival, initiation, and progression of tumor cell ( Figure 1A ; Chen et al, 2019 ; Lan et al, 2019 ; Du et al, 2020 ; Huang et al, 2020 ; Nombela et al, 2021 ).…”
