Lysosomal-associated Transmembrane Protein 4B (LAPTM4B) Decreases Transforming Growth Factor β1 (TGF-β1) Production in Human Regulatory T Cells

Abstract: Background: TGF-␤1 produced by regulatory T lymphocytes suppresses excessive immune responses. Results: Lysosomal-associated transmembrane protein 4B (LAPTM4B) was found to inhibit TGF-␤1 production in Tregs. Conclusion: LAPTM4B, known to exert oncogenic functions in tumor cells, also plays a role in the immune system. Significance: LAPTM4B may represent a new therapeutic target to modulate immunosuppression by Tregs.

“…encoding TCA intermediates in various solid tumors(LaGory & Giaccia 2016;Lee et al 2016;Pollard et al 2005). With reference to LAPTM4B, it has been extensively studied for genotyping studies associated with various cancers from serum as well as investigated for its expression in PBMCs and platelets(Cheng et al 2016;Gnatenko et al 2010;Huygens et al 2015;Ma et al 2015;Mo et al 2014).The current study confirmed the elevated expression of HIF-1α in peripheral blood of solid tumor patients as compared to normal healthy controls that have very low or negligible expression of HIF-1α. The stabilization of HIF-1α leads to the overexpression of hypoxia responsive genesthe MDR1.…”

“…LAPTM4B plays a number of oncogenic roles, and high levels of this protein are associated with poor prognosis in many types of cancers. LAPTM4B was recently identified as binding to GARP and serving as a negative regulator for TGF-b1 production in human Tregs (29). Tregs also express high levels of FAS molecules (Fas cell surface death receptor, also known as APO-1/CD95), which play an important role in CD95L-mediated apoptosis (30).…”

“…Previous studies have proven that GARP is necessary for Tregs to activate latent TGF-β1 [ 17 18 ]. Thus, we explored whether GARP is also expressed on MSCs and whether it is important for MSCs to inhibit T cells through activating latent TGF-β1.…”
