Lysis of allogeneic and autologous melanoma cells by IL-7-induced lymphokine-activated killer cells

Abstract: SmmuaryIn order to assess the potential of interleukin 7 (IL-7) as an immunotherapeutic agent in human melanoma, we have evaluated the in vitro activity of IL-7-induced lymphokine-activated killer (LAK) cells from patients with advanced melanoma against allogeneic and autologous melanoma cells. Peripheral blood lymphocytes (PBLs) from 14 patients with stage III melanoma were isolated and incubated in the presence of 1,000 U ml-' IL-7 and 1 U ml-' IL-2 for comparison. LAK-ell activity was determined by a 24 h c… Show more

“…Human peripheral blood lymphocytes (PBL) were isolated from a healthy donor (M.B.) by Ficoll-Hypaque centrifugation as described previously (Bö hm et al, 1994) and were stimulated with 20 U per mL IL-2 or 1000 U per mL g-IFN for 4 h (both from R&D Systems).…”

“…In vitro, IL-7 is used to support APCinduced T-cell priming with a higher specificity than in the presence of IL-2 (Kos and Millbacher, 1992;Celis et al, 1994). Compared with other cytokines, anti-tumour response induced by paracrine-secreted IL-7 seems to be more strictly dependent on T cells (Hock et al, 1993;Miller et al, 1993), although IL-7 is able to induce LAK activity (Alderson et al, 1990;Bohm et al, 1994), to enhance NK activity (24) and to generate tumoricidal activity in monocytes (Alderson et al, 1991). In a comparative study in a mouse tumour model using a murine plasmocytoma cell line, the IL-7-secreting gene-modified tumour cells showed, in contrast to other cytokine-secreting tumour cells, a dependency of CD4+ T cells on early tumour rejection, whereas CD8+ T cells were required for long-term tumour eradication (Hock et al, 1993).…”

“…Furthermore, a difference in the percentage of lysis between before vaccination (B) and 2 weeks after the third vaccination (A) of Ͼ10% was thought to be necessary for the difference to be considered significant, as tested with PBMCs obtained from healthy donors. 21 Based on these criteria, an enhancement of spontaneous lytic activity against autologous melanoma cells was detected in the PBMCs of 7 of 12 evaluable patients (patients 1, 7, 9, 1b, 2b, 3b, and 6b), whereas a reduction in spontaneous lytic activity in the PBMCs of 3 of 12 patients (patients 5, 8, and 10) was observed after vaccination. Spontaneous lytic activity against the allogeneic melanoma cell line was found to be enhanced in 6 of 12 patients and increased in 4 of 12 patients after vaccination (Fig 2).…”
