Lysine Succinylation and Acetylation in Pseudomonas aeruginosa

Abstract: Pseudomonas aeruginosa is a multi-drug-resistant human opportunistic pathogen largely involved in nosocomial infections. Unfortunately, effective antibacterial agents are lacking. Exploring its physiology at the post-translational modifications (PTMs) level may contribute to the renewal of combat tactics. Recently, lysine succinylation was discovered in bacteria and seems to be an interesting PTM. We present the first succinylome and acetylome of P. aeruginosa PA14 cultured in the presence of four different ca… Show more

“…Most notably, Gingipain R1 (B2RM93) harbors 11 Ksuc modification sites. The distribution of the number of Ksuc sites per protein is different from that found in E coli , M tuberculosis , P aeruginosa , and V parahaemolyticus , all of which have only one Ksuc site in 40%‐50% of identified proteins …”

“…All 233 identified Ksuc proteins account for 11.1% of the total proteins in P gingivalis ATCC 33277. The percentage of Ksuc proteins to total proteins is similar to that identified in other bacterial species, including Escherichia coli , Mycobacterium tuberculosis , Pseudomonas aeruginosa , and Vibrio parahemolyticus . Of the 233 identified Ksuc proteins, approximately 75% (174) possess a single Ksuc site and 11.6% (27) contain 3 or more Ksuc sites.…”

“…Several proteins involved in oxidative stress response and antibiotic sensitivity exhibited a modified expression level. The abundance of three proteins (PA14_18690, OpdB, and OprG) were increased and seven others (PA14_47550, PA14_18330/50, ObgE, MexG, MexH, and OmpD) were reduced with both treatments as compared to the control (Aendekerk, 2005;Breidenstein et al, 2008;Dotsch et al, 2009;Balasubramanian et al, 2013;Verstraeten et al, 2015;Sakhtah et al, 2016;Chevalier et al, 2017;Gaviard et al, 2018;Wolloscheck et al, 2018). Several other protein levels showed differences between the two lactonase treatments.…”

“…Succinyl CoA is also a precursor for protein succinylation, and such posttranslational modification is used to fine-tune the metabolism (Yang et al, 2015). In addition to proteins involved in TCA and gluconeogenesis, four proteins necessary for alginate biosynthesis, namely AlgU, MucB, AlgR, and AlgC, were found to be succinylated in P. aeruginosa (Gaviard et al, 2018).…”
