Lymphocyte Subpopulations in Homozygous Sickle Cell Anaemia

Abstract: Patients with sickle cell disease have an increased tendency to develop frequent and severe pyo-genic infections. Lymphocyte subpopulations were studied in 14 patients with homozygous sickle cell anaemia (SCA) using the OKT monoclonal antibody. The mean absolute lymphocyte counts observed in patients with SCA were similar to that of controls. The OKT8+ lymphocytes were significantly higher in SCA patients than in controls. The mean OKT3+ and OKT4+ lymphocytes were significantly lower in SCA patients than in co… Show more

“…These results are in agree ment with previous reports which demonstrated simi lar results in S C A [15,16,29] and after splenectomy [26,27,31]. Since the spleen contains approximately 50% o f B lymphocytes [31], its absence may determine a marked redistribution o f these cells among other compartments.…”

“…These results show that the number of lymphocytes was increased in S C A and in splenectomized subjects as have been previously reported [15,16,[24][25][26][27][28][29][30]], This increment is more pronounced in S C A and the inter pretation for this finding is unclear. Differences be tween the effect of autosplenectomy and surgical splenectomy and the influence of previous transfu sions should be considered.…”

“…In particular, studies of cellular immunity and humoral factors have yielded conflicting results [3, 4]. Also, the few reports of the roles and pattern of cytokines have focused mainly on American patients [15, 16, 17]; studies of Arab patients with the milder disease have not been published.…”

“…Studies of neutrophil function and humoral immunity have yielded inconsistent findings [2, 3]. While helper T (CD4+) cells and cytotoxic/suppressor (CD8+) T cells appear to be quantitatively normal in children with the disease, abnormalities have been reported in adult patients [4, 5, 6]. There is abundant evidence that CD4+ and CD8+ cells can be further divided into subsets by their function and pattern of cytokine secretion.…”

“…These re sults might be expected, since high numbers of B lym phocytes [24][25][26][27][28] and a normal percentage of CD4+ T cells [26,27], whose helper activity is required for the antibody response to PWM, have been found in the peripheral blood of patients with SCD. As stated in the Introduction, patients with SS hemoglobinopathy showed a normal production of antibody after anti genic stimulation by the intradermal, subcutaneous, or intramuscular routes [18][19][20], whereas the response to intravenous inoculation of antigen was depressed [9,21].…”
