Am J Physiol Regulatory Integrative Comp Physiol 282: R1104-R1112, 2002 10.1152/ ajpregu.00075.2001.-This study investigated the capacity of conjugated linoleic acids (CLA) to reduce ex vivo antigeninduced release of eicosanoids in a type I hypersensitivity model. Guinea pigs were fed a diet containing 0.25% safflower oil (control) or 0.25% CLA [43% trans (t)10, cis (c)12; 41% c9, t11/t9, c11 18:2] for 2 wk before and during sensitization to ovalbumin (OVA). Lungs, tracheas, and bladders were incubated in physiological saline solution (PSS) for 1 h (basal mediator release) and challenged with OVA (0.01 g/l PSS) for 1 h (mediator release in response to antigen). Eicosanoids were quantified by HPLC/tandem mass spectrometry or enzyme immunoassay. CLA feeding resulted in no change in basal release but decreased eicosanoid release from sensitized tissues in response to antigen challenge in the following manner: thromboxane B 2, 6-keto-prostaglandin (PG)F1␣, PGF 2␣, PGD2, PGE2 by 57-75% in lung, 45-65% in trachea, and 38-60% in bladder; and leukotriene C 4/D4/E4 by 87, 90, and 50% in lung, trachea, and bladder, respectively. We previously showed that dietary CLA lowered prostaglandin (PG)E 2 and histamine release in response to antigen challenge (42) in a guinea pig model for type I (immediate) hypersensitivity, a standard model for studying asthma and allergies (12, 41). The mechanism by which CLA lower PGE 2 levels may be, at least in part, inhibition of cyclooxygenase (COX), the enzyme responsible for the initial steps in converting arachidonic acid into prostanoids [PG and thromboxanes (TX)] (2). CLA isomers are similar to linoleic acid (the precursor of arachidonic acid), except the double bonds are in a conjugated formation (i.e., a 1,3-diene, not methylene interrupted). This structural difference (a conjugated diene) in similar 20-C fatty acids has been shown to inhibit the enzymatic activity of COX in vitro as measured by PG production (28).For many years, prostanoids have been implicated in the pathogenesis of type I hypersensitivity disorders such as asthma. The present study was designed to compare the conjugated linoleic acid (CLA) isomeric distribution pattern in the liver of suckling rats in relation to those in the milk and maternal diet. Silver-ion HPLC was used to separate individual CLA isomers. It was found that the isomeric distribution pattern in the milk was very similar to that in the maternal dietary fat. However, the CLA isomeric distribution patterns in the liver phospholipids (PL) and triacylglycerols were different from those in the diet and milk. In the liver PL, total cis/trans isomers accounted for 63·6-63·9 % of total CLA, which was in contrast to the values of 88·1-89·1 % in the milk and diet. In the liver PL, total trans/trans isomers were 20·6-20·8 % of the total CLA isomers whereas they were only 2·6-3·7 % in the milk and diet. It is concluded that trans/trans-CLA were preferentially incorporated into the liver whereas for the incorporation of cis/trans-CLA there was partial discrimination.

Lipids are essential components of living organisms and support multiple biological and physiological functions in the human body. They serve as the structural building material of all membranes of cells and organelles. Lipids and their derivatives also serve as signaling molecules that facilitate a variety of physiological functions. In addition, lipids are recognized as important biomarkers of disease and are involved in several pathological conditions. Individual fatty acids can influence biological processes, such as membrane‐associated receptors and signal transduction systems, and ion channels. Recent literature also demonstrates a specific role of fatty acids in gene modulation and protein expression to influence risk of chronic disease. Polyunsaturated fatty acids (PUFAs) are the focus of substantial research interest in this area mainly because of their ability to be converted to various bioactive regulatory molecules, such as prostaglandins, leukotrienes, and thromboxanes. Arachidonic acid (AA, 20:4n‐6) and eicosapentaenoic acid (EPA, 20:5n‐3) are two major players in the regulation and production of these bioactive molecules. In addition, an absolute requirement of docosahexaenoic acid (DHA) is necessary for normal neural and retinal development in the infant and young child. Besides the study of these PUFA molecules, numerous animal and human studies also suggest that other dietary lipids, such as cholesterol and saturated fatty acids, increase serum cholesterol as well as LDL‐cholesterol concentrations. In summary, this chapter provides a contemporary understanding of food lipids in nutrition and health, especially the underlying cellular and molecular mechanisms of the actions of these bioactive dietary lipids.
