Lymphatic growth factors are expressed in human gingiva and upregulated in gingival fibroblasts after stimulation

Abstract: The localization of VEGF-C, -D, and VEGFR-3 expression imply that signaling via VEGFR-3 is linked to vascular homeostasis and keratinocyte function under normal conditions in gingiva. Inflammatory stimulation of HGFs upregulates VEGF-C and -A expression and may contribute to angiogenesis and lymphangiogenesis.

“…The HGFs were isolated and cultured as previously described. 24 All HGFs were cultured in α-minimum essential medium (α-MEM; Hyclone, Logan, UT, USA) containing 10% FBS (Gibco, Grand Island, NY, USA) at 37°C with 5% CO 2 in a cell incubator.…”

“…In a study on sheep injected with radioactive albumin in gingiva, as much as 64.7% of the injected protein was recovered in the prescapular and cervical lymph vessels 7.5 h after injection (15). The gingiva is well supplied with lymphatic vessels (7,16,17), and when lymph is formed, muscle movements and soft tissue deformation may promote a rapid transport of fluid via the lymphatic system. This is in contrast to the bone‐embedded interstitial tissue in the mandibular canal, and may explain why we observed a much slower washout rate from this compartment.…”

“…After challenging with lipopolysaccharide (LPS) or IL-6/sIL-6R complex, gingival fibroblasts increased the secretion and gene expression of VEGF-A and -C compared to non-induced controls, while VEGF-D was not detected. Altogether, VEGF-A and -C contributes to both the angiogenesis and lymphangiogenesis processes in the pathogenesis of periodontitis [109].…”
