Lymphangiogenesis: fuel, smoke, or extinguisher of inflammation’s fire?

Abstract: Lymphangiogenesis is a recognized hallmark of inflammatory processes in tissues and organs as diverse as the skin, heart, bowel, and airways. In clinical and animal models wherein the signaling processes of lymphangiogenesis are manipulated, most studies demonstrate that an expanded lymphatic vasculature is necessary for the resolution of inflammation. The fundamental roles that lymphatics play in fluid clearance and immune cell trafficking from the periphery make these results seemingly obvious as a mechanism… Show more

“…Заметим, что функция лимфатических коллекторов в процессе воспаления не сводится лишь к транспорту антигенов и иммунокомпетентных клеток: эндотелиоциты лимфатических капилляров способны регулировать активность и фенотип иммуноцитов. В различных условиях лимфангиогенез может как тормозить, так и усугублять течение воспалительных процессов [26].…”

“…The growth of lymphatic vessels or lymphangiogenesis is actively involved in multiple pathological conditions, including tissue and organ inflammation [32]. In particular, lymphangiogenesis could be advantageous in decreasing inflammation related to lymphedema [24].…”

“…The lymphatic capillaries of the small intestine are fenestrated. In addition, they are surrounded by a fragmented basement membrane with anchoring filaments attached at the abluminal place[24]. Intestinal lymphatic pathophysiology in cholestatic rats could be related to inflammation, with increased Th1 and Th2 lymphocyte activation in the small bowel and in the mesenteric lymph complex of microsurgical extrahepatic cholestatic rats[26].The drainage function of lymphatic vessels is crucial for inflammation in the intestinal interstitium.…”

“…The molecular and cellular mechanisms of lymphangiogenesis involved in organ graft rejection, cancer metastasis, and lymphedema are discussed in detail elsewhere. Moreover, lymphangiogenesis has an active and complex role in pathological circumstances such as inflammation, possibly including allergies and asthma . A common factor in these diverse lymphangiogenic‐related conditions is activation of the VEGF pathway, which includes ligands (VEGF‐C, VEGF‐D), receptors (VEGFR2 and VEGFR3), intracellular signaling enzymes (PI3K, mitogen‐activated protein kinases), and transcription factors (Prox‐1).…”
