Lyb-8.2: A new B cell antigen defined and characterized with a monoclonal antibody

Abstract: A DBA/1 B10.D2-specific monoclonal antibody (CY34) is described which defines a new murine B lymphocyte differentiation antigen designated Lyb-8.2. The ontogeny, strain distribution, and cell-surface density of the antigen were studied by radioimmunoassay and by fluorescence-activated cell sorter (FACS) analysis. Lyb-8.2 appears to be expressed on pre-B cells and on all mature B lymphocytes. Lyb-8.2 molecules immunoprecipitated from surface labeled B10.D2 spleen cells migrated in polyacrylamide gels with an ap… Show more

“…38 Mouse CD22 (mCD22) is detected in the cytoplasm early in B-cell development, is absent from the surface of newly emerging IgM ϩ B cells, is present at a low density on the immature B220 lo IgM hi B cells, and is fully expressed by mature B220 hi IgD ϩ B cells of the BM. 39,40 In the periphery, mCD22 is expressed at high levels on all B-cell subsets including follicular and marginal zone B cells and peritoneal B1 cells. However, a minor subset of immature B cells in the spleen recently derived from BM, expresses low density CD22.…”

“…The purified mouse anti-human IgG monoclonal antibody (mAb) R10Z8E9 (34) was kindly provided by Professor R. Jefferis and Dr. M. Goodall and is available from Recognition Systems (University of Birmingham Science Park, Birmingham, United Kingdom). The hybridoma CY34.1.2, which produces the mouse (IgG1) anti-mouse CD22 antibody CY34 (35), was obtained from the American Type Culture Collection (Rockville, MD).…”

“…The CY34 Epitope Includes Portions of Domains 1 and 2-CY34 is an allele-specific mouse anti-mouse CD22 mAb (35) that has been reported to bind the CD22 BALB/c allele, but not the DBA/2J allele (55). The CD22 C57Bl allele identified in the present study also binds CY34 (28).…”

“…In the BXD RI lines, this variation in the 908 structural gene was found to cosegregate with the Coh, Gpi-1, and Tam-i loci on chromosome 7 (Table 1). A recently identified B-lymphocyte antigen, Lyb-8.2, is encoded by a gene, Lyb-8, that maps to this region also (18). We have called the 908 structural gene RP2, with the designations RP2b and RP2d for alleles carried by strains C57BL/6J and DBA/2J, respectively.…”
