Ly-6I, a New Member of the Murine Ly-6 Superfamily with a Distinct Pattern of Expression

Abstract: A new member of the mouse Ly-6SF, designated Ly-6I, has been isolated as a gene homologous to a segment of the Ly-6C gene. A single allelic difference in the mature protein sequence was identified, which is similar to other Ly-6SF members. Ly-6I mRNA has been detected in a wide range of tissues and cell lines, and a rabbit polyclonal Ab has been used to determine that Ly-6I protein is present at a low constitutive level on cell lines from several different lineages. In contrast to Ly-6C and Ly-6A/E, the Ly-6I … Show more

“…Although the role of Ly6i is unknown, it has been proposed as a maturation marker for T and B lymphocytes as well as for subsets of monocytes and granulocytes (44). The Immunoresponsive gene1 (Irg1) was highly up-regulated as well.…”

“…CD11b is expressed on macrophages, granulocytes, NK cells, and dendritic cells. Sca-1 (Ly-6A/E) is constitutively expressed on hematopoietic stem cells and is also upregulated in peripheral lymphocytes and monocytes upon activation in BALB/c mice (27,33). One of the most studied aspects of the Ly-6 family of proteins, and particularly Ly-6A/E, is their exquisite sensitivity to induction by IFN-␣/␀ and IFN-␄ (20,39).…”

“…CD11b is widely expressed on myeloid lineage cells, including macrophages and neutrophils, as well as on NK cells (15,24). Sca-1 is expressed on all hematopoietic stem cells and is also an activation marker of diverse types of lymphoid and myeloid lineage cells (23,31,33,40). In PBS-treated mice, 6.6% of splenocytes were CD11b Ï© / Sca-1 ÏȘ (Fig.…”

“…Ly-6A and Ly-6E were recognized to be alleles of the same gene, and the Ly-6A/E protein is readily detected by the Sca-1 mAb and is perhaps the most useful marker in the isolation of the long term reconstituting hemopoietic stem cell (9,10). Recently, the gene for Ly-6I was characterized (11) and was found to be near the genes for both Ly-6A/E and TSA-1/Sca-2/Ly-6e on mouse chromosome 15 (12,13).…”
