Lung Targeted Lipopolymeric Microspheres of Dexamethasone for the Treatment of ARDS

Abstract: Acute respiratory distress syndrome (ARDS), a catastrophic illness of multifactorial etiology, involves a rapid upsurge in inflammatory cytokines that leads to hypoxemic respiratory failure. Dexamethasone, a synthetic corticosteroid, mitigates the glucocorticoid-receptor-mediated inflammation and accelerates tissue homeostasis towards disease resolution. To minimize non-target organ side effects arising from frequent and chronic use of dexamethasone, we designed biodegradable, lung-targeted microspheres with s… Show more

“…The release of DTX was studied from pristine DTX, DPLGA nanoparticles, and ADN-DPLGA nanoparticles by suspending in a Float-A-Lyzer (G2, Spectrum, Repligen, MA, USA) in two different release media, namely phosphate buffer saline pH 7.4 and sodium acetate buffer pH 5.0 containing Tween 80 to maintain sink condition and facilitate release [ 37 ]. The tests were performed at 37 °C (n = 6).…”

“…The release of efinaconazole was studied by suspending efinaconazole-loaded spanlastics in a Float-A-Lyzer (G2, Spectrum, Repligen, MA, USA) in phosphate buffer saline pH 7.4. The tests were performed at 37 °C (n = 6) by introducing the dialyzers into 100 mL release media stirred at 100 rpm [ 24 ]. Efinaconazole released at different time intervals up to 4 h was analyzed using the above-mentioned method by withdrawing 0.5 mL of release media.…”

“…The focus of the research has always been the use of nano-carriers for aerosol delivery that will eventually lead to the improved therapeutic efficacy of the drug [4]. However, being a treatment of vital organs, the material-borne toxicity of such carriers should not be overlooked [5].…”
