Lung cancer stem cells—characteristics, phenotype

Abstract: Lung cancer remains a major cause of cancer-related deaths worldwide with unfavourable prognosis mainly due to the late stage of disease at presentation. High incidence and disease recurrence rates are a fact despite advances in treatment. Ongoing experimental and clinical observations suggest that the malignant phenotype in lung cancer is sustained by lung cancer stem cells (CSCs) which are putative stem cells situated throughout the airways that have the potential of initiating lung cancer formation. These c… Show more

“…Lung adenocarcinoma and bronchoalveolar carcinoma has been associated with stem cells from the bronchoalveolar duct junction region. Studies on mouse models demonstrated the association of Matrixmetalloproteinase-10 (MMP10) with the maintenance and tumorigenicity of lung CSC (31,32).…”

“…CD44, principally, CD44v isoforms are CSC markers that play essential roles in the execution of the fundamental features of CSCs [19, 26, 101, 102]. Hence, CD44 has been proposed to be used for the isolation and enrichment of CSCs in lung cancers including MPM [22, 24, 26, 100, 103]. …”

“…reported that CSCs were enriched in multiple phenotypically distinct subpopulations [20]. Such conflicting results are likely attributable to the heterogeneous origins and high plasticity of lung CSCs [8, 9]. Considering the inconsistencies in the markers identified for lung CSCs, it is preferable to define CSCs based on their biological functions.…”

“…It is now widely accepted that this heterogeneity is generated mainly by cancer stem cells (CSCs), which have both tumour-initiating ability and differentiation potential [5, 6]. Several studies have reported the isolation and characterization of lung CSCs, and several putative markers for lung CSCs have been identified, including CD133, CD44, CD166, aldehyde dehydrogenase (ALDH) and side population (SP) phenotypes [79]. However, controversies and uncertainties remain, and no consensus markers for lung CSCs have yet been identified [7].…”
