Luminescent lanthanide nanoparticle-based imaging enables ultra-sensitive, quantitative and multiplexed in vitro lateral flow immunoassays

Abstract: We developed a portable, fast, highly sensitive and quantitative in vitro assay for on-site biomolecule detection by combining the remarkable optical properties of new lanthanide-doped nanoparticle probes with a simple reader coupled to a smartphone.

“…This multiplexed assay was 100 times more sensitive than the comparative AuNP-based LFA, reaching LoDs in the pg mL –1 range for all the biomarkers investigated. Additionally, the researchers developed a portable and cost-effective smartphone-based readout system (see section ; Figure Bii) …”

“…The maturation stage of these new tools will probably soon open them the doors of commercial applications. In particular, the bio-analytical fields are already tackling the use of Ln-NPs for in vitro applications such as lateral flow immunoassays, 286 ELISA like detection, 287 or luminescence microscopy. 288 However, the high sensitivity obtained with Ln-NPs is based on the development of dedicated instruments, working with a time-delayed detection mode, that will require dedicated time-resolved readers to unravel the best of the sensitivity of these nano-objects.…”

“…21 Vanadate is considered as an excellent optical entity to enhance the global luminescent emission because of its large absorption cross-sections and effective substrate sensitization band. 22,23 Moreover, Eu 3+ ions in vanadate matrix have been reported to emit strong and stable luminescence. 24 The vanadate NPs doped with Eu 3+ yielded the typical red luminescence of Eu 3+ , which can be excited with near-UV light through an energy transfer from the vanadate anion.…”
