Hsa_circ_0003645 Promotes Breast Cancer Progression by Regulating miR-139-3p/HMGB1 Axis

Abstract: Background: The aim of the present study was to investigate the effect of over-expressing circular RNA (circ_0003645) on cell functions and its molecular mechanism in breast cancer. Methods: The expression profile of circ_0003645, breast cancer cell lines, and the transcription levels of circular RNA, miRNA and HMGB1 gene were detected by qRT-PCR. Flow cytometry analysis was manipulated to evaluate cancer cell proliferation and cell apoptosis. The correlation between miR-139p-3p and circular_0003645 or HMGB1 w… Show more

“…Mechanistically, cirCHIPK3 serves as a sponge for miR-193a, thereby reactivating HMGB1/PI3K/AKT signaling to promote proliferation, migration and invasion of breast cancer cells 69 . Another circRNA, Hsa_circ_0003645 have also been proved that can regulate miR-139-3p/HMGB1 axis to promote breast cancer progression 70 . However, there is a paucity of reports on how RNA sponges, or even competing endogenous RNA (ceRNA) regulatory networks, impact HMGB1-mediated breast cancer progression.…”

“…evidence confirmed that circRNAs serve as biomarkers in diverse clinical diseases, [8] including breast cancer. [9][10][11] For instance, hsa_-circ_0001785 [12] and hsa_circ_0003645 [13] were reported to act as biomarkers and participate in the advancement of breast cancer. A current study verified that hsa-circRNA-0088088 was considerably expressed in s both serum exosomes and tissues, which might function as competing endogenous RNAs (ceRNAs) and play a crucial role in the advancement of breast cancer.…”

“…For example, the overexpression of circ-ABCB10 and circ_001783 were found to facilitate BC progression through sponging miR-1271 and miR-200c-3p respectively [26,27]. In addition, circ_0003645 was showed to promote BC proliferation by targeting miR-139-3p/HMGB1 signaling axis [28], and circ_0001667 could enhance BC cell survival by regulating TAZ in Hippo signal pathway [29]. In our study, we also demonstrated the upregulation of CircPRMT5 in BC tumors and cell lines, and the silencing of CircPRMT5 suppressed the aggressive phenotype of BC cells.…”
