Exosomes Derived from Mesenchymal Stem Cells Protect the Myocardium Against Ischemia/Reperfusion Injury Through Inhibiting Pyroptosis

Abstract: Objective Mesenchymal stem cells (MSCs) show unique advantages in cardiomyocyte repairment. Exosomes derived from MSCs can enhance the viability of myocardial cells after ischemia/reperfusion (I/R) injury and regulate inflammation response. The study was designed to ascertain whether MSCs-exo protect the myocardium against I/R injury through inhibiting pyroptosis, and the underlying mechanisms. Methods and Results Experiments were carried out in H/R and I/R model. Cell … Show more

“…They hypothesized that hucMSC-derived exosomes may protect cardiomyocytes from apoptosis by regulating the expression of the Bcl-2 family and promote the tube formation and migration of vascular endothelial cells ( 68 ). Exosomes secreted by hucMSCs protect cardiomyocytes from anoxia-reoxygenation injury ( 69 ). Similar studies are equally effective in mouse ischemia/reperfusion injury models ( 70 ).…”

“…And pyroptosis plays a pivotal role in the pathogenesis of various cardiovascular diseases, especially MI [23]. Suppressing NLRP3-mediated pyroptosis pathway protects myocardial ischemia-reperfusion injury [24][25][26]. In our study, ColCaNPs could help colchicine attenuate cardiac pyroptosis, and at the same time, colchicine-containing calcium carbonate nanoparticles signi cantly inhibited the expression of TLR4, NFκB and NLRP3 (Fig.…”

“…The mechanism by which MSCs and secretome inhibit pyroptosis has been more deeply studied in other disease models. Several new findings showed that MSCs exosomes inhibited NLRP3 expression and pyroptosis of cardiomyocytes and myocardial infarction by delivering miRNA-320b or long non-coding RNA (lncRNA) KLF3-AS1 (Mao et al, 2019;Tang et al, 2020). Liu et al (2020c) found that cotransplantation of a chitosan thermosensitive hydrogel with BMSCs contributed to a better outcome in myocardial infarction, which is manifested by the inhibition of inflammatory response and alleviation of pyroptosis in vascular endothelial cells.…”
