Lsm Proteins Are Required for Normal Processing and Stability of Ribosomal RNAs

Abstract: Depletion of any of the essential Lsm proteins, Lsm2-5p or Lsm8p, delayed pre-rRNA processing and led to the accumulation of many aberrant processing intermediates, indicating that an Lsm complex is required to maintain the normally strict order of processing events. In addition, high levels of degradation products derived from both precursors and mature rRNAs accumulated in Lsm-depleted strains. Depletion of the essential Lsm proteins reduced the apparent processivity of both 5 and 3 exonuclease activities in… Show more

“…This could be the reason why correctly processed U14 is targeted to the foci. In contrast, U3 is not bound by Nop1p, Nop56p, and Nop58p before it reaches the final maturation stages at the nucleolar body (Kufel et al, 2000;Kufel et al, 2003). Assuming that trafficking of U3 snoRNA to the nucleolus occurs independently of Nop1p binding, U3 can escape sequestration in the foci.…”

“…LSM4 and LSM8 (both Class I) are best characterized in yeast, where they form complexes involved in RNA splicing and processing [18,19,214]. They are predicted to have similar roles in metazoan cells, but there function in mammals is not well described, in part because the LSM4 null mouse is embryonic lethal [215].…”

“…Indeed overexpression of LSM5 stabilized box H/ACA snoRNAs, which were specifically depleted from the synthetic lethal strain, and presumably thereby rescued the synthetic lethality. Although depletion of Lsm proteins affects the stability of pre-rRNA and rRNAs, it does not impact snoRNAs (49). Therefore, the effect of LSM5 overexpression on the stability of box H/ACA snoRNAs was likely indirect and could have been mediated via Srp40p, thus closing the chain between SHM2 and SRP40.…”
