LRP6 downregulation promotes cardiomyocyte proliferation and heart regeneration

Abstract: The adult mammalian heart is thought to be a terminally differentiated organ given the postmitotic nature of cardiomyocytes. Consequently, the potential for cardiac repair through cardiomyocyte proliferation is extremely limited. Low-density lipoprotein receptor-related protein 6 (LRP6) is a Wnt co-receptor that is required for embryonic heart development. In this study we investigated the role of LRP6 in heart repair through regulation of cardiomyocyte proliferation. Lrp6 deficiency increased cardiomyocyte ce… Show more

“…To achieve optimal parathyroid hormone (PTH) signalling activation, LRP6 may possibly act as a coreceptor for the PTH receptor [ 72 ]. Cardiomyocyte proliferation is regulated by LRP6 via the ING5/P21 pathway [ 73 ]. Therefore, further molecular and functional investigations are required to fully understand these possible LRP6 interactions, which must be taken into consideration when Lrp6 is targeted for a potential therapeutic purpose.…”

“…Intriguingly, the accumulation of β-catenin has been observed upon constitutive activation of ERBB2 signaling, specifically mediating cardiomyocyte dedifferentiation (102). Ablation of lipoprotein-related receptor protein LRP6 (a coreceptor interacting with Frizzled receptor in Wnt/β-catenin signaling) in infarcted mouse hearts stimulates robust regenerative processes through the proliferation of pre-existing cardiomyocytes via a β-catenin independent mechanism, involving ING5 (inhibitor of growth family member 5)/p21 (206).…”

“…Nuclei were determined as tetraploid if their intensity values were within 1.5-2.5 times range of the reference cells. Polyploidy was assigned if DAPI intensity was greater than 2.5 times range of reference cells, as previously described 53 .…”
