In the present study, release properties of antibiotic-loaded cement-type nanocomposites of biomimetic apatite and calcium sulfate were studied. Nanocrystalline component of the nanocomposite was synthesized by soaking a mixture of calcium phosphate reactants in tris-buffered simulated body fluid (SBF). The release patterns of cephalexin and gentamicin from both pure calcium sulfate and nanocomposite cements into SBF were collected up to 144 h and fitted by Higuchi and Weibull equations. The effect of loaded antibiotics on physical properties of the cements was also evaluated. Fast release behavior of both antibiotics was obtained from calcium sulfate matrix, in which 80-85% of the loaded antibiotics were liberated during the first 10 h of elution. In contrast, an administered elution was acquired from nanonocomposite materials so that the release was controlled, in all cases, by a combined mechanism; major mechanism was drug diffusion through the matrix and the minor was matrix dissolution. The results showed that the initial setting time and injectability of cements were increased from 7 min and 71% for pure calcium sulfate cement (powder-to-liquid ratio = 2.5 g/mL) to 33 min and 95% for the nanocomposite cement containing 60 wt % apatite, respectively. The compressive strength of nanocomposite was about 0.9 MPa, nearly four times lower than that of pure calcium sulfate. In addition, the use of cephalexin monohydrate did not influence the setting time and compressive strength of the cements, whereas (adding) gentamicin sulfate significantly improved these properties.

Powder-based inkjet three-dimensional printing (3DP) to fabricate pre-designed 3D structures has drawn increasing attention. However there are intrinsic limitations associated with 3DP technology due to the weak bonding within the printed structure, which significantly compromises its mechanical integrity. In this study, calcium sulphate ceramic structures demonstrating a porous architecture were manufactured using 3DP technology and subsequently post-processed with a poly (ε-caprolactone) (PCL) coating. PCL concentration, immersion time, and number of coating layers were the principal parameters investigated and improvement in compressive properties was the measure of success. Interparticle spacing within the 3DP structures were successfully filled with PCL material. Consequently the compressive properties, wettability, morphology, and in vitro resorption behaviour of 3DP components were significantly augmented. The average compressive strength, Young's modulus, and toughness increased 217%, 250%, and 315%, following PCL coating. Addition of a PCL surface coating provided long-term structural support to the host ceramic material, extending the resorption period from less than 7 days to a minimum of 56 days. This study has demonstrated that application of a PCL coating onto a ceramic 3DP structure was a highly effective approach to addressing some of the limitations of 3DP manufacturing and allows this advanced technology to be potentially used in a wider range of applications.

Abstract-The introduction of additive manufacturing (AM), often referred to as three-dimensional (3D) printing, has initiated what some believe to be a manufacturing revolution, and has expedited the development of the field of biofabrication. Moreover, recent advances in AM have facilitated further development of patient-specific healthcare solutions. Customization of many healthcare products and services, such as implants, drug delivery devices, medical instruments, prosthetics, and in vitro models, would have been extremely challenging-if not impossible-without AM technologies. The current special issue of the Annals of Biomedical Engineering presents the latest trends in application of AM techniques to healthcare-related areas of research. As a prelude to this special issue, we review here the most important areas of biomedical research and clinical practice that have benefited from recent developments in additive manufacturing techniques. This editorial, therefore, aims to sketch the research landscape within which the other contributions of the special issue can be better understood and positioned. In what follows, we briefly review the application of additive manufacturing techniques in studies addressing biomaterials, (re)generation of tissues and organs, disease models, drug delivery systems, implants, medical instruments, prosthetics, orthotics, and AM objects used for medical visualization and communication.
