Low M r Phosphotyrosine Protein Phosphatase Associates and Dephosphorylates p125 Focal Adhesion Kinase, Interfering with Cell Motility and Spreading

Abstract: Low M r phosphotyrosine protein phosphatase interferes in vivo with the activation of several growth factor receptors and is transiently redistributed, following cell stimulation with platelet-derived growth factor, from the cytosol to the cytoskeleton. We demonstrate here that this phosphatase also participates in the regulation of cell spreading and migration, pointing to its involvement in cytoskeleton organization. Low M r phosphotyrosine protein phosphatase-overexpressing fibroblasts are, indeed, less spr… Show more

“…Activation of FAK, overexpressed in several human cancers, induces survival, proliferation, and motility of cells in culture (32). In our study, FAK phosphorylation of Tyr 397 in COUP-TFIItransfected A549 cells was increased, mainly in cells grown on fibronectin-coated plates.…”

“…Indeed, H 2 O 2 has been shown to inactivate LMW-PTP through the oxidation of cysteine residues (76). Interestingly, LMW-PTP has been shown to dephosphorylate FAK (30,77). Another substrate of LMW-PTP is EphA2 (78,79).…”

“…Although the identity of the oxidase remains unknown, DPI sensitivity and the requirement for the Rac insert region suggest the participation of a Nox-family member. Interestingly, LMWPTPase also dephosphorylates p120FAK, inhibiting cell spreading (Rigacci et al, 2002); thus the control of FAK phosphorylation is another regulatory target for ROS produced during cell spreading (Chiarugi et al, 2003).…”
