PURPOSE:To determine the influence of low-level laser therapy on femoral growth plate in rats. METHODS:Thirty male Wistar rats aged 40 days were divided into two groups, G1 and G2. In G1 the area of the distal growth plate of the right femur was irradiated at one point using GaAlAs laser 830 nm wavelength, output power of 40 mW, at an energy density of 10 J/cm 2 . The irradiation was performed daily for a maximum of 21 days. The same procedure was done in G2, but the probe was turned off. Five animals in each group were euthanized on days 7, 14 and 21 and submitted to histomorphometric analysis. RESULTS:In both groups the growth plate was radiographically visible at all moments from both craniocaudal and mediolateral views.On the 21st day percentage of femoral longitudinal length was higher in G2 than G1 compared to basal value while hypertrophic zone chondrocyte numbers were higher in G1 than G2. Calcified cartilage zone was greater in G1 than in G2 at all evaluation moments.Angiogenesis was higher in G1 than in G2 at 14th and 21st days. CONCLUSION:The low-level laser therapy negatively influenced the distal femoral growth plate.Key words: Lasers. Bone Development. Chondrocytes. Laser Therapy, Low-Level. Rats. RESUMO OBJETIVO:Determinar a influência do Laser Terapêutico de Baixa Potência sobre a placa de crescimento de ratos. MÉTODOS:Trinta ratos Wistar machos com 40 dias de idade foram divididos em dois grupos, G1 e G2. O grupo G1 foi submetido à irradiação com laser GaAlAs 830 nm, potência de saída de 40 mW, e densidade de energia de 10 J/cm2. A irradiação foi aplicada diariamente por um período máximo de 21 dias. O mesmo procedimento foi realizado no grupo G2, com a probe desativada. Cinco animais em cada grupo foram sacrificados nos dias 7, 14 e 21 e submetidas à análise histomorfométrica. RESULTADOS:Em ambos os grupos, o disco fisário esteve radiograficamente visível em todos os momentos nas incidências craniocaudal e médio-lateral. No 21º dia a porcentagem de comprimento longitudinal do fêmur foi maior em G1 que em G2 em relação ao valor basal, e o número de condrócitos da zona hipertrófica foi maior em G1 que em G2. A zona de cartilagem calcificada estava maior em G1 em relação a G2 em todos os momentos de avaliação. A angiogênese foi maior em G1 que em G2 nos 14º e 21º dias. Low-level laser irradiation (LLLI) can enhance stem cell (SC) activity by increasing migration and proliferation. This study investigated the effects of LLLI on proliferation, enzymatic activity, and growth factor production in human umbilical cord mesenchymal SCs (hUC-MSCs) as well as the underlying mechanisms. hUC-MSCs were assigned to a control group (non-irradiation group) and three LLLI treatment groups (635 nm group, 808 nm group, and 635/808 nm group). Laser power density and energy density of 20 mW/cm2 and 12 J/cm2, respectively, were used for each experiment. The proliferation rate was higher in the 635 nm as compared to the other groups. LLLI at 808 nm did not induce cell proliferation. ROS levels in cells exposed to 635, 808, and 635/808 nm radiation were increased by 52.81%, 26.89%, and 21.15%, respectively, relative to the control group. CAT, tGPx, and SOD activity was increased. LLLI at 808 nm increased the levels of IL-1, IL-6, and NFκB but not VEGF. LLLI improved hUC-MSCs function and increased antioxidant activity. Dual-wavelength LLLI had more potent effects on hUC-MSCs than single-wavelength treatment. LLLI has potential applications in the preconditioning of hUC-MSCs in vitro prior to transplantation, which could improve the regenerative capacity of cells.

IntroductionThe nerve fiber or axon, is responsible for transmitting information between neuron cells. 1 In humans, axons are generally on the scale of 1 μm thick and some are more than 1 m long, 1 and in the peripheral nerve system (PNS), they are either unmyelinated or myelinated by Schwann cells.2 These cells are mainly wrapped around axons and form a multi-layered sheath preparing myelin structures for axons. 2,3 In case of injury, Schwann cells are activated and play a key role in nerve repair mechanism, not only concerning Wallerian degeneration and remyelination in the injured position, but also in promoting axonal growth. 4 Wallerian degeneration occurs when an axon is cut or injured, in which the axon degenerates distal to the injury site. [5][6][7] This is followed by secondary phenomena such as breakdown of the myelin sheet and activation of the Schwann and macrophages that clear up the debris. 8,9Nerve injuries are divided into 3 basic categories of neuropraxia, axonotmesis and neurotmesis based on the extent of injury, pathology and physiology according to Sir Herbert Seddon's scheme, 5,6 neuropraxia, the mildest nerve injury, is a temporary loss of sensory function. 10The injured site usually fully recovers within few weeks or months, and no Wallerian degeneration occurs in this type of injury. Axonotmesis is when axons are more seriously damaged and interrupted. 10 In this type of injury, the surrounding epineurium and perineurium layers remain almost intact, but there could be only partial recovery depending on the extent of injury to the axons. In addition, Wallerian degeneration may occur distal to Methods:In this research work, the effect of LLLT (780 nm) on the regeneration process and reconstruction of injured peripheral right side sciatic nerve was investigated. Twelve adult male Wistar rats underwent surgery in aseptic conditions under general anesthesia to induce a lesion to their right side sciatic nerve according to standard protocol. Before suturing the location, only the experimental group was treated by laser. The damaged nerve was directly irradiated with (2 J, 100 mW, 40 seconds). The irradiation procedure was terminated in 21 days with little improvement (4 J, 200 mW, 40 seconds) across the skin surface of experimental group. Rats were selected randomly from each group to be sacrificed on different periods and histopathological examination was carried out on the extracted nerves.Results: Significant acceleration of revascularization and angiogenesis of the injury site was observed in the experimental group. Furthermore, a reduction of hemorrhages and increase in blood supply was observed. Also, Wallerian degeneration decreased while higher axonal density compared to the control rats was observed. Moreover, the cross-section analysis of the injured area on the 14th and 21st days as post-surgery showed that the nerve sheath diameter in the lesion area of the experimental group was reduced.
