Low Field NMR Determination of pKa Values for Hydrophilic Drugs for Students in Medicinal Chemistry

Abstract: For an interdisciplinary approach on different topics of medicinal and analytical chemistry, we applied a known experimental pKa value determination method on the field of the bench top nuclear magnetic resonance (NMR) spectrometry of some known biologically active pyridine-based drugs, i.e., pyridoxine hydrochloride, isoniazid, and nicotine amide. The chemical shifts of the aromatic ring protons in the 1 H NMR spectrum change depending on the protonation status. The data were analyzed on dependence of the che… Show more

“…Unlike Nam and nicotinic acid (NA) which are chemically stable, NR + , NMN and NAD + are readily hydrolyzed to Nam, with their rapid decomposition particularly promoted by heat and basic aqueous solutions (pH ≥ 8). Under non-buffered conditions, the hydrolysis of the glycosidic bond leads to the release of Nam, which possesses a p K a at 3.35 [47]. The release of Nam results in the in situ generation of pyridinium hydroxide, resulting in further NR + hydrolysis.…”

“…Nevertheless, high-purity samples with concentrated parent compounds are required for the NMR measurement due to the low sensitivity (Malet-Martino and Holzgrabe, 2011). The routinely applied nuclei for the measurement of PPCPs mixtures include 1 H, 13 C, 19 F, and 31 P (Gladden et al, 2012;Malet-Martino and Holzgrabe, 2011;Zivkovic et al, 2017). Among them, 1 H is the most sensitive one for monitoring the reaction process (Malet-Martino and Holzgrabe, 2011).…”

“…Identification of natural amino acids consists of using increment calculations, multiplicity predictions and chemical shifts in order to identify an unknown amino acid (Zivkovicet al, 2017). Another experiment consists of logP determination, where signal integration is used for the calculation of lipophilicity (Soulsby and Chica, 2017;Zivkovic et al, 2018). Furthermore, for the purpose of understanding the relative signal integration, the experiment where the quantitative analysis of multicomponent mixtures of over-the-counter (OTC) pain killer drugs as an example for nonsteroidal anti-inflammatory drugs (NSAIDs) is determined (Zivkovicet al, 2017).…”
