Low extracellular magnesium does not impair glucose-stimulated insulin secretion

Abstract: There is an increasing amount of clinical evidence that hypomagnesemia (serum Mg 2+ levels < 0.7 mmol/l) contributes to type 2 diabetes mellitus pathogenesis. Amongst other hypotheses, it has been suggested that Mg 2+ deficiency affects insulin secretion. The aim of this study was, therefore, to investigate the acute effects of extracellular Mg 2+ on glucose-stimulated insulin secretion in primary mouse islets of Langerhans and the rat insuli… Show more

“…3B and S3). TRPM7 channel control of β-cell insulin secretion is also exemplified by a recent report showing that TRPM7 channel knockdown in rat insulinoma cells (INS-1) augments insulin secretion [41]. Taken together, these data suggest that TRPM7 channels not only play an important role in -cell development, but also in limiting insulin secretion.…”

“…This is not surprising given that expression of TRPM7 is almost ubiquitous. In mouse islets, expression of TRPM7 is eight times higher than that of TRPM6, which is the other Mg 2+ channel present in the mouse islets [110]. In human β-cells TRPM6 is not expressed.…”

“…Deficiency of TRPM7 reduces total cellular Mg 2+ at least in some cells [111]. Knockdown of Trpm7 in INS-1 cells by siRNA increases insulin secretion in response to glucose [110]. It appears that TRPM7 plays a role in mediating glucose-induced insulin secretion possibly by regulating the cytoplasmic Mg 2+ concentration.…”
