Low-density lipoprotein receptor-mediated endocytosis of PEGylated nanoparticles in rat brain endothelial cells

Abstract: Poly(methoxypolyethyleneglycol cyanoacrylate-co-hexadecylcyanoacrylate) (PEG-PHDCA) nanoparticles have demonstrated their capacity to diffuse through the blood-brain barrier after intravenous administration. However, the mechanism of transport of these nanoparticles into brain has not yet been clearly elucidated. The development of a model of rat brain endothelial cells (RBEC) in culture has allowed investigations into this mechanism. A study of the intracellular trafficking of nanoparticles by cell fractionat… Show more

“…Phenylarsine oxide has been reported to be an inhibitor of clathrin-mediated endocytosis [23]. Filipin blocks the caveolae-mediated process in non-specific adsorptive endocytosis [24]. Colchicine is known to inhibit macropinocytosis [25], which provides non-specific endocytosis of macromolecules, whereas cationic poly-L-Lysine acts as an uptake inhibitor for cationic delivery systems.…”

“…The medium was removed and the cells were pretreated with PhAsO (10 mmol/L), filipin (5 mg/mL), colchicine (10 mmol/L), excessive Lf (100 mmol/L), and polylysine (400 mg/mL) for 10 mins at 371C. PhAsO is reported to inhibit clathrindependent endocytosis (Visser et al, 2004), and filipin is known to block the caveolae-mediated process (Kim et al, 2007). Colchicine is an inhibitor of macropinocytosis (Liu and Shapiro, 2003), whereas cationic polylysine could inhibit the uptake of cationic vectors.…”

“…For the detailed set-up of the DPD simulation, the readers are referred to our previous studies [71,109]. For the model of NPs, we consider that the NP is formed by a rigid core with a surface coated polymer [110]. A typical model system will be the PEGylated Au NPs, with the core and shell representing Au NP and PEG polymer, respectively.…”
