Loss of the High-Affinity Pentamidine Transporter Is Responsible for High Levels of Cross-Resistance between Arsenical and Diamidine Drugs in African Trypanosomes

Abstract: Treatment of many infectious diseases is under threat from drug resistance. Understanding the mechanisms of resistance is as high a priority as the development of new drugs. We have investigated the basis for cross-resistance between the diamidine and melaminophenyl arsenical classes of drugs in African trypanosomes. “…T. brucei brucei bloodstream forms of strain Lister 427 were cultured at 37°C and under a 5% CO 2 atmosphere exactly as described previously (Gudin et al, 2006) in HMI-9 media (Invitrogen, Paisley, UK) supplemented with 10% fetal bovine serum (FBS) (BioSera, Ringmer, UK).
