Loss of adult skeletal muscle stem cells drives age-related neuromuscular junction degeneration

Abstract: Neuromuscular junction degeneration is a prominent aspect of sarcopenia, the age-associated loss of skeletal muscle integrity. Previously, we showed that muscle stem cells activate and contribute to mouse neuromuscular junction regeneration in response to denervation (Liu et al., 2015). Here, we examined gene expression profiles and neuromuscular junction integrity in aged mouse muscles, and unexpectedly found limited denervation despite a high level of degenerated neuromuscular junctions. Instead, degenerated… Show more

“…2) While NMJs acquire age-related features, the underlying cause of such changes remains debated. In particular, it remains unknown whether NMJs are a focal site of pathology or degenerate due to deleterious changes elsewhere in skeletal muscles, motor neurons or other cells [1316]. 3) The rate of aging among NMJs located in different muscles, within the same muscle, and even in the same motor unit varies considerably [17].…”

“…Max‐projection z‐stack images of NMJs were generated with ImageJ software. The post‐synaptic side was identified based on the entry of the terminal axon and as the concave side of the NMJ [3,4].…”

“…Whole EDL muscle contractility and force generation were analyzed using an ASI muscle contraction system (Aurora Scientific, Aurora, Canada) as described previously [3,4]. Briefly, mice were anaesthetized and TA muscles removed.…”
