Loop-Mediated Isothermal Amplification Detection of SARS-CoV-2 and Myriad Other Applications

Abstract: As the second year of the COVID-19 pandemic begins, it remains clear that a massive increase in the ability to test for SARS-CoV-2 infections in a myriad of settings is critical to controlling the pandemic and to preparing for future outbreaks. The current gold standard for molecular diagnostics is the polymerase chain reaction (PCR), but the extraordinary and unmet demand for testing in a variety of environments means that both complementary and supplementary testing solutions are still needed. This review hi… Show more

“…Previously, only four primers were included in ACTB primer set to detect ACTB gene, which prolonged the reaction (Huang et al, 2020 ; Poon et al, 2005 ). The reaction time was significantly shortened by adding loop primers (Avelar, 2019 ; Jamwal et al, 2021 ; Moore et al, 2021 ; Zhang et al, 2020 ). Therefore, we designed a new set of primers, ACTB‐n, which also detected the ACTB sequence with the additional loop primers.…”

“…Faced with the challenge of the SARS-CoV-2 pandemic, laboratories worldwide have been applying a variety of strategies and methods to meet the exceptionally high testing demand (18, 19). For molecular testing, these include rRT-PCR performed directly on crude samples without nucleic acid extraction (20)(21)(22)(23)(24), isothermal ampli cation techniques (25,26) and CRISPR-based nucleic acid detection assays (27)(28)(29). In our approach, we maintained a standard rRT-PCR testing work ow and the associated high detection sensitivity and speci city by combining three pragmatic and straightforward measures to stepwise increase our testing capacity, generating several options to exibly adapt sample throughput according to the current demand.…”

“…However, unlike LAMP, RPA often produces non-specific amplification even in water controls, an effect that has been reported by the original inventors of the technology ( 54 ). Finally, it should be noted that RPA kits are currently commercialized by a single company, TwistDx [now part of Abbott Laboratories, US ( 66 )] which has full control over the price and availability of the kits. RPA has not yet been approved by the food and drug administration (FDA), and for now is intended for research-only applications ( 65 , 67 ).…”
