Longitudinal study of specific antibodies to toluene diisocyanate TDI-human serum albumin HSA conjugate in patients with TDI-induced asthma

Abstract: Background :An appreciable number of patients with toluene diisocyanate (TDI)-induced asthma have high serum levels of specific IgE (sIgE) antibody to TDI-human serum albumin conjugate (HSA). A recent investigation suggested a role of specific IgG (sIgG) in the development of TDI asthma. Methods :We observed the changes in the levels of specific IgE and IgG antibodies to TDIHSA conjugate in TDI-induced asthmatic patients during seven years avoidance. Results :Six subjects with high sIgE and five with high sIgG w… Show more

“…It has been estimated that, after cessation of workplace exposure, the plasma half-life of detectable IgE antibody specific for the chemical allergen is in the range of 4 months to 2 years. 85,93 Likewise, it was observed that measurements of specific IgE antibody were more likely to be positive if blood samples were drawn from subjects within 30 days of the last exposure to the chemical allergen. Finally, the significance of IgE antibody in isocyanate-induced asthma has been challenged by the work of Jones et al 94 The authors found a striking absence, within the bronchial mucosa, of RNA message for Ce, e heavy chain, and IL-4, the cytokine that promotes the B lymphocyte switch to IgE synthesis, in patients sensitized to diisocyanates after specific inhalation challenge.…”

“…IgEs are labile in serum with a half-life measured in days. Despite this fact, serum levels of antibody against a unique, pointed allergen challenge (without repeated challenge) have a half-life of 3-7 years (16,17) due to the long life of antibody-producing plasma cells. In addition, persons exhibiting high levels of IgE early in life are highly prone to developing adult atopic allergies (particularly respiratory), indicating that a point measurement of IgE in adulthood is not an unreasonable measure of historical IgE levels (18 -20), a point supported by our data (Table 4).…”

“…In our previous study, we have shown that the level of serum-specific IgE to DI-HSA is significantly higher in DI-OA patients compared with unexposed healthy controls. 38 Serum IgE levels are usually elevated in asthmatic patients, adversely affecting asthma pathogenesis. 39 In vitro experiments have shown that DI can induce oxidative stress in the lungs and affect the function of lung tissue.…”
