Longitudinal Study of Cellular and Systemic Cytokine Signatures to Define the Dynamics of a Balanced Immune Environment During Disease Manifestation in Zika Virus–Infected Patients

Abstract: Understanding the host immune response during Zika virus (ZIKV) infection will provide valuable insights into ZIKV immunopathogenesis. In this study, characterization of cellular changes and immune mediators of ZIKV-infected patients was performed, allowing for the identification of key infection-associated markers.

“…Specifically, IgG1 and IgG3 were the subclasses induced following ZIKV infection, closely resembling DENV-infected patients. 35 Moreover, none of the ZIKV patients in our study displayed severe symptoms to suggest occurrence of antibodydependent enhancement (ADE), 30 and similar observations were also reported from Brazil. This observation is also supported by another study, in which the profiles of ZIKV-neutralising antibodies of patients from Nicaragua, Sri Lanka and Thailand were not affected by previous DENV infection.…”

“…Anti-ZIKV IgM and IgG levels of ZIKV patients from the Singapore outbreak in 2016 [28][29][30] were longitudinally assessed using virion-based ELISA. 18,[24][25][26][27] The majority of the patients showed a robust ZIKVspecific humoral response (Figure 1a IgG isotypes produced by ZIKV patients were then determined, and highest titres of anti-ZIKV IgG1 and IgG3 subtypes were produced at early convalescent for IgG3 and late convalescent for IgG1 (Figure 1d).…”

“…Nevertheless, a series of histologic, immunologic and transcriptomic analyses were used to determine the impact of ZIKV infection on pregnancy progression and the cellular responses of the host towards the infection. All three babies were born at term with no observable birth defects, and all of the ZIKV‐infected mothers were clinically well, as with majority of ZIKV‐infected patients . However, different degrees of infection were apparent in the level of CD14 + monocytes between the three patients.…”

“…As confirmed by MFA of immune‐phenotyping and cytokine data, the three pregnant patients can be easily separated based on their immune response. Importantly, by comparing their cytokine profiles to those of the healthy non‐pregnant female controls, several immune mediators such as IL‐1RA, EGF, RANTES and IP‐10 were associated with the acute phase of ZIKV infection, a finding that was also observed in ZIKV‐infected non‐pregnant patients . Interestingly, EGF and IP‐10 are typically expressed at higher levels in women carrying a foetus with developmental anomalies .…”
