Long-Term Sensitization in Aplysia : Biophysical Correlates in Tail Sensory Neurons

Abstract: A fundamental problem in the cellular analysis of learning and memory is the identification of the neuronal substrates of long-term information storage and their relation to short-term cellular alterations. In this report, biophysical correlates of long-term sensitization of a simple withdrawal reflex in the mollusc Aplysia were examined. A voltage-clamp analysis of the sensory neurons that control the reflex, 24 hours after sensitization training, revealed a significant reduction in net outward current. The r… Show more

“…Isolated ganglia were treated for 1.5 hr with 5 ~M 5-HT (Sigma, St. Louis, MO) and then frozen, essentially as described in Noel et al (1991) but without [35S]methionine. Sensitization training consisted of four blocks of electrical shocks over 1.5 hr delivered to the posterior body wall (Scholz and Byrne 1987; W.L. Lee, M. Aguirre, L.J.…”

“…To further evaluate the potential role of these proteins in long-term sensitization and facilitation, we measured mRNA changes in sensory neurons following behavioral training. Animals received four blocks of shocks over 1.5 hr to one side of the body as previously described (Scholz and Byrne 1987; W.L. Lee, M. Aguirre, L.J.…”

“…Whereas a single tail shock induces short-term memory (STM) for sensitization lasting minutes, repeated spaced shocks produce LTM for sensitization lasting days to weeks (Frost et al 1985;Scholz and Byrne 1987;Castellucci et al 1989;Goldsmith and Byrne 1993;Cleary et al 1998;Levenson et al 2000;Sutton et al 2001a;see also, Carew et al 1971;Pinsker et al 1973). Recently, we distinguished a third phase of memory for sensitization, intermediate-term memory (ITM), that is induced by repeated spaced shocks and lasts 1-3 h after training (Sutton et al 2001a).…”

“…The reflex exhibits sensitization, a simple form of learning, in which responses elicited by weak test stimuli are augmented by training with strong, usually noxious, stimuli (Carew et al 1971;Pinsker et al 1973;Scholz and Byrne 1987). Short-term sensitization (lasting from seconds to minutes) relies on covalent modification of pre-existing proteins.…”
