Long-Term Pharmacokinetics of Transdermal Testosterone Gel in Hypogonadal Men1

Abstract: Transdermal delivery of testosterone (T) represents an effective alternative to injectable androgens. Transdermal T patches normalize serum T levels and reverse the symptoms of androgen deficiency in hypogonadal men. However, the acceptance of the closed system T patches has been limited by skin irritation and/or lack of adherence. T gels have been proposed as delivery modes that minimize these problems. In this study we examined the pharmacokinetic profiles after 1, 30, 90, and 180 days of daily application o… Show more

“…There are androgen receptors throughout the cortex and subcortical regions in the human brain though which T administration affects behaviors (Chawnshang, 2002). Published pharmacokinetics show that T levels peak 18 h after Androgel ® administration (Swerdloff et al, 2000). In order to confirm that T levels were actually raised (i.e.…”

“…The testosterone administration experiment used a 10 g dose of a topical hydrophilic drug called Androgel ® (1 percent testosterone gel) prescribed for hypogonadal men (Swerdloff et al, 2000). “…The daily doses vary between 5 and 10 g, each delivering 5 to 10 mg testosterone. Relatively steady levels of serum testosterone are obtained after a few days [20]. One potential problem is transferal of androgen via close contact.…”
