“…The current results were not obtained from an effective intervention, but rather from the observational significant and clinically relevant associations between weight loss and clinical and structural OA development. 13,14 A limitation of our validation of the compartment-accumulated cavity score is that the score does not show to what extent the quantification corresponds to few larger lesions or multiple smaller lesions, or whether focal defects such as fibrillations or fissures contribute to the score. Therefore, further validation could be performed against more invasive scorings from optical coherence tomography 15 or from histopathology.…”

“…miR‐146a fl/+ mice have been backcrossed with WT (C57BL/6J) mice for over seven generations to maintain in a C57BL/6J background. Using the strategy described previously to achieve cartilage‐specific overexpression of miRNA (de Vos et al., 2017), they were bred with Col2a1‐Cre mice to generate miR‐146aTG mice Col2a1Cre;miR‐146a [B6.Cg‐Tg(Col2a1‐Cre;miR‐146afl]. Genomic DNA isolated from the tail was analyzed by PCR using the primers used for confirmation of proper homologous recombination and genotyping are listed in Table S1.…”

“…GlcN is largely used for osteoarthritis at an average dose of 1,500 mg per day but its usefulness in other pathologies has not been explored in clinical trials. Whereas, long term treatment seems to delay the progression of knee arthritis (28), multiple studies have shown no superiority of glucosamine vs. placebo (89), no improvement of cartilage damage (90) and no role in prevention of osteoarthritis in overweight women (91). Moreover, at the usual doses, GlcN may induce an increase in intraocular pressure (92).…”
