Long‐term effect of lithium on the uptake of 5‐hydroxytryptamine by human platelets.

Abstract: 1The content, uptake and storage of 5-hydroxytryptamine (5-HT) in platelets were determined in eight manic-depressive patients not on lithium (Li); in ten manic-depressive patients on Li; and in ten apparently normal persons as controls. 2 Platelets from the patients, whether or not on Li, contained less 5-HT than platelets from normal people. 3 5-HT uptake and storage by platelets from untreated patients were significantly lower than those of control subjects. 4 When patients had been on Li for 3 months or lo… Show more

“…These results are also in accord, as previously discussed elsewhere (Carli et al, 1997a), with previous studies showing diminished 5-HT uptake in platelets from patients with affective disorders (Coppen et al, 1980;Healy et al, 1983;Meltzer et al, 1981;Tuomisto and Tukiainen, 1976). Indeed, it has been proposed that the chronic administration of Li 1 could help to compensate by elevating the deficient uptake, as shown in patients under lithemic therapy (Born et al, 1980;Coppen et al, 1980;Meltzer et al, 1983;ThiesFlechtner et al, 1994). Also, Abouh-Saleh et al (1992) suggested that patients with enhanced 5-HT uptake, as a result of chronic administration of Li 1 , show a decreased morbidity when compared to patients that had a lower uptake.…”

“…The reported range for normal subjects is large (12). Most studies have employed platelet-rich plasma (PRP) for estimation (21)(22)(23). 5-HT uptake estimated by this method may be confounded by the presence of protein and other chemicals present in plasma.…”

“…Effects of lithium on 5-HT uptake in platelets were demonstrated also in man (Murphy et al, 1969;Born et al, 1980;Coppen et al, 1980;Corona et al, 1982).…”

“…Murphy et al (1969) and Born et al (1980) showed that chronic lithium treatment up to three months increased the serotonin uptake in blood platelets of manic-depressive patients. Coppen et al (1980) reported an increased uptake ofplatelets 5-HT after lithium treatment of six weeks, one year, and after lithium prophylaxis of 4.3 years on average.…”
