Long-term administration of tacrolimus and everolimus prevents high cholesterol-high fructose-induced steatosis in C57BL/6J mice by inhibiting de-novo lipogenesis

Abstract: AimTo investigate the effects of tacrolimus (TC) and everolimus (EV) on non-alcoholic steatohepatitis (NASH) induced by high fat, high cholesterol and fructose (fast food) diet in C57BL/6J mice.Materials and MethodsC57BL/6J mice were divided into four groups (n=8). 1) Standard Chow (SC); 2) Fast food (FF) diet; 3) FF + Tacrolimus (TC, 1mg/kg) and; 4) FF + Everolimus (EV, 1mg/kg) and treated for 16 weeks. Serum and tissue samples were analyzed for evidence of inflammation, fibrosis, lipogenesis, and apoptosis.R… Show more

“…[ 15 ] Previous studies in rodents have shown that fructose can facilitate de novo lipogenesis. [ 16 17 18 ] Feeding rats with fructose increased hepatic levels of upstream regulators of de novo lipogenesis (i.e., sterol regulatory element-binding protein 1c [ SREBP-1c ] and carbohydrate response element-binding protein [ ChREBP ]) and key lipogenic enzymes (i.e., fatty acid synthase [FAS], acetyl-CoA carboxylase [ACC], and stearoyl-CoA desaturase-1 [SCD-1]). To verify these findings at a cellular level, in the present study, we incubated HepG2 cells with fructose or palmitic acid (PA) to investigate their effects on different sources of accumulated lipid.…”

“…Recent studies have reported that a prolonged diet of high fructose combined with high fat induces steatohepatitis, fibrosis and severe liver damage in mice (Leung et al, 2016 ; Baena et al, 2017 ; Love et al, 2017 ). While mice administered only liquid fructose did not show hypertriglyceridemia, a combination of high fat and high fructose induced TG accumulation, diminished insulin signaling, and increased cholesterol deposition.…”
