Long Noncoding Competing Endogenous RNA Networks in Pancreatic Cancer

Abstract: Pancreatic cancer (PC) is a highly malignant disease characterized by insidious onset, rapid progress, and poor therapeutic effects. The molecular mechanisms associated with PC initiation and progression are largely insufficient, hampering the exploitation of novel diagnostic biomarkers and development of efficient therapeutic strategies. Emerging evidence recently reveals that noncoding RNAs (ncRNAs), including long ncRNAs (lncRNAs) and microRNAs (miRNAs), extensively participate in PC pathogenesis. Specifica… Show more

“…Previous literature has shown that expression of lncRNAs is altered in many malignancies including pancreatic cancer. 27 It has been reported that H19 expression is high in pancreatic cancer tissues. It has also been shown that H19 expression is higher in metastasized tumors.…”

“…On one hand, lncRNAs act as miRNA sponges and competitive endogenous RNAs (ceRNAs) of miRNAs to hinder the binding of miRNAs to target genes. 21 LncRNA HAGLR sponges miR-338-3p to accelerate 5-Fu resistance in gastric cancer via targeting the LDHA-glycolysis pathway. 22 The ceRNA network formed by lncRNA–miRNA–mRNA plays a crucial part in several cancers including gastric cancer.…”

“…This phenomenon leads to the deregulation of miRNA function with several effects on the genome. Modulation of PVT1 that takes part in pancreatic carcinogenesis and chemoresistance to gemcitabine, is also considered a potent druggable target, while it is proven that the knockdown of PVT1could suppress the autophagy mechanism and limit the chemoresistance to gemcitabine by miR-143/HIF-1α/VMP1 axis [ 114 , 115 , 116 , 117 , 118 , 119 , 120 ]. Another example of miRNA-related autophagy inhibition that suppresses PDAC grow this mediated by miR-7 and miR-372 [ 121 , 122 ].…”
